| A reversed-phase LC-MS-MS method was developed for simultaneous determination of dexamethasone acetate (DXA), betamethasone (BM), prednisolone (PR), triamcinolone acetonide (TCA) in cosmetics treating acne or for skin whitening. The procdure was optimized as: Column - Phenomenex Gemini C-18 (150 x 4.6 mm, 5μm); Mobile phase - ACN-H2O in gradient mode; Sample injection volume - 5μl; Electrospray ionnization source was operated in positive mode and multi reaction monitoring (MRM) mode using the transition of m/z 435.20 " 397.25; m/z 393.11 " 147.06; m/z 361.22 "147.13; m/z 345.20 "397.25 and m/z 375.18 "161.17 were used to quantify DXA, BM, PR, TCA and methyl prednisolone (IS), respectively. Validated in accordance to AOAC guidelines, the method proved to meet the requirements: Wide linearity range (20-750ppb) and high correlation coefficient (R2> 0.9982), very good accuracy (recovery rate - 90.2– 112.2%) and intra-day precision (RSD < 6.5%), typical LOD, LOQ values (0.1-0.3ppb and 0.33-9.9ppb for each of all four glucocorticoids). On practical application, 20 commercial cosmetic products for acnes and whitening skin Tra Vinh province was controlled to determine these four corticoids. Of these cosmetics, 35% were detected to be adulterated with corticosteroids. |
