A reverse-phase HPLC method was developed for determination of rosmarinic acid and luteolin in leaf extracts of Perilla frutescens (L.) Britt. The method was established as: Column - Zorbax SB C18 (25 cm × 4.6 mm, 5 μm); Detector - UV (300nm); Mobile phase: phosphoric acid 0.1% - acetonitrile (70-30); flow rate: 10µg.mL-1 for rosmarinic acid and 1.9 – 12.6µg.mL-1 for luteolin (r > 0.999): good intra – and inter-day precision variations (RSD < 3.0%), and desirable mean recovery for both rosmarinic acid (99.07 ± 0.53 %, RSD = 0.69 %) and luteolin (100.09 ± 1.25 %, RSD = 1.63 %). In lack of any current official monograph on quality control of Perilla leaf extracts, the proposed method proved practically applicable for the purpose.
