Tóm tắt An isocratic liquid chromatography coupled with tandem mass spectrometry (LC-MS/MS) was developed for simultaneous assay of metronidazole (MET) and spiramycin I (SPIY I) in human plasma. The blank human plasma with MET, SPY I, and roxithromycin (ROX) added as was internal standards was treated by protein precipitation with acetonitrile. The analytes were chromatographed as follows: Column - Phenomenex NX C18 column (100 mm x 3.0 mm; 5.0 µm); Mobile phase - acetonitrile and ammonium formate 5 mM pH 7.4 (80:20, v/v); Flow rate - 0.4 mL/min.; Injection volume - 3 µL; Column temperature - 40oC; Detector – MS in positive ion electrospray ionization (ESI+) with multi-reaction monitoring (MRM) mode; m/z 171.75 → 128.10 for MET, 422.25 → 174.20 for SPY I, and 837.50 → 679.45 for ROX. Linearity covered the concentration range of 0.050-25.235 and 0.025-12.725 µg/ml for MET and SPY I, respectively. The intra- and inter-day accuracy and precisions (CV) were from 95.40 % to 106.39 % with CV value below 15.0 % for both MET and SPY I. The method showed appliable to determination of pharmacokinetic parameters of MET and SPY I in human plasma.
